You are following a Master in the field of Analytical Chemistry, Pharmaceutical Sciences, Life Sciences, or any other relevant studies. Further you:
- Are available for a minimum of 9 months

The Bacterial Vaccines department is currently developing adjuvanted vaccine formulations. It is critical
to assess the quality and stability of those adjuvants alone and after mixing with antigens to create a candidate drug product. One of the focus areas of the Analytical Development team is to develop analytical tools that provide insight into the interaction between antigens and adjuvants and study the impact on the drug product formulation. Analysis of a nanoparticulate adjuvant system in combination with glycoconjugates and proteins is a major challenge in the field of vaccines but at the same time critical to understand immunogenic potential and efficacy of a vaccine 1,2. Routinely, we use an array of various biophysical techniques to characterize those formulations. In the current project, we plan to develop a set of methods to characterize liposomal and/or oil-in-water adjuvant formulations.
- Implement an AF4-MALS (Asymmetric-Flow Field-Flow Fractionation- Multi Angle Light Scattering) protocol that was developed externally
- Then, optimize the protocol for the antigen-adjuvant combinations that are relevant for our current candidate bacterial vaccine compositions.
- Test resolution of antigens, aggregates, fragments and adjuvants.
- Assess whether the method can be used to investigate interactions between antigens and adjuvants.
The Bacterial Vaccines department, part of Johnson & Johnson Innovative Medicine, is located in Leiden
Bio-Science Park (Archimedesweg 6, 2333 CN). Bacterial Vaccines has well-equipped laboratories and collaborates with other departments in Leiden, such as Vaccine Process & Analytical Development (VPAD) and Quality Control (QC), which provide expertise to Bacterial Vaccines and its external partners for the development of their prototype vaccines.
