Lipid-linked oligosaccharides act as precursors to glycoconjugate vaccines during bioconjugation. The
analysis of these molecules can serve as a powerful approach for the discovery of novel antigens as well as
further developing our understanding of the polysaccharide portion of the glycoconjugate vaccine.
Therefore, it is essential to develop suitable, accurate and robust techniques that can efficiently characterize
lipid-linked oligosaccharides (1, 2). This internship will focus on developing an end-to-end liquid
chromatography – mass spectrometry workflow for polysaccharide characterization which will help drive
the discovery and development of novel vaccine antigens.

  • Plan, execute and write a research report on your own project.
  • Explore and optimize polysaccharide purification and fluorescent labeling techniques in order to create a universal workflow with high labeling efficiency.
  • Optimize a liquid chromatography – mass spectrometry workflow for the analysis of polysaccharides.
  • Application of the workflow to different vaccine antigens

The Bacterial Vaccines department, part of Johnson & Johnson Innovative Medicine, is located in Leiden
Bio-Science Park (Archimedesweg 6, 2333 CN). Bacterial Vaccines has well-equipped laboratories and
collaborates with other departmentsin Leiden, such as Vaccine Process & Analytical Development (VPAD)
and Quality Control (QC), which provide expertise to Bacterial Vaccines and its external partners for the
development of their prototype vaccines.
